Ribonucleotide reductase of rabbit bone marrow.

چکیده

Ribonucleotide reductase activity was increased in extracts of bone marrow from rabbits treated with methyl phenyldiazenecarboxylate, a hemolytic agent which results in an increased number of reticulocytes in blood. The enzyme has been partially purified from bone marrow of rabbits treated with this agent. Enzymatic reduction of cytosine ribonucleotides requires a dithiol, magnesium ions, and ATP and is stimulated by ferrous ions. CDP is a better substrate than CTP or CMP. The purified extracts can be separated into two protein fractions which must be combined for activity, indicating that the enzyme system consists of more than one component.

Phase I study of GTI-2040, an antisense to ribonucleotide reductase, in combination with high-dose cytarabine in patients with acute myeloid leukemia.

PURPOSE Inhibition of ribonucleotide reductase reduces the availability of the endogenous pool of deoxycytidine and may increase cytarabine (AraC) cytotoxicity. We performed a phase I dose escalation trial of AraC combined with GTI-2040, a 20-mer antisense oligonucleotide shown in preclinical studies to decrease levels of the R2 subunit of ribonucleotide reductase, to determine the maximum tole...

Investigation of solvent effect on the active site energy of Carbonic Anhydrase and Ribonucleotide Reductase

Enzymes catalyze many biological reactions. The rates of chemical reaction in the presence ofenzymes are, in some cases, accelerated more than 10 orders of magnitude relative to thecorresponding rates in solution. In this paper a comparison between optimized structures of two enzyme molecules in aspect ofenergy and dipole moment in different conditions including presence of metallic ion, without...

Objective- To investigate the effect of intratendinous injection of bMSCs on the rate and extent of tendon healing after primary repair in a rabbit model. Design- Experimental study. Animals- Twenty seven skeletally mature New Zealand white rabbits weighing 1.8- 2.5 kg were used. Twenty rabbits were used as the experimental animals, and seven others were used as a source of bone marrow-derived ...

MRI evaluation of rabbit bone marrow after acute irradiation

Background: Magnetic Resonance Imaging (MRI) is a safe modality and useful in characterizing normal and abnormal bone marrow (BM). MRI also presents a more global view of BM than biopsy therefore, it may provide a better understanding of hematologic disorders. The purpose of this study was to monitor radiation-induced alterations of BM in acute phase of irradiation (1-10 day after total body ir...

Short-term treatment with novel ribonucleotide reductase inhibitors Trimidox and Didox reverses late-stage murine retrovirus-induced lymphoproliferative disease with less bone marrow toxicity than hydroxyurea.

We evaluated the ability of a short course of treatment with the ribonucleotide reductase (RR) inhibitor hydroxyurea (HU) and two novel RR inhibitors Trimidox (TX) and Didox (DX) to influence late-stage murine retrovirus-induced lymphoproliferative disease. LPBM5 murine leukaemia virus retrovirus-infected mice were treated daily with HU, TX or DX for 4 weeks, beginning 9 weeks post-infection, a...
